Role of Nitric Oxide in the TRPV1-mediated Melanogenesis

Abstract

|
We investigated the possible role of transient receptor potential vanilloid 1 (TRPV1) in melanogenesis in B16melanoma cells. Capsaicin (CAP), a TRPV1 agonist, increased production of melanin and nitric oxide (NO) in a dosedependentmanner, which was inhibited by capsazepine (CPZ), a TRPV1 antagonist. The CAP-induced melanin synthesiswas blocked by L-NAME and cPTIO, an inhibitor of NO synthase and a NO scavenger, respectively. CAP increasedintracellular Ca2+ concentration and BAPTA/AM, an intracellular Ca2+ chelator, inhibited the CAP-induced production ofNO and melanin. These results suggest that TRPV1 may stimulate melanogenesis through intracellular Ca2+-mediatedproduction of NO.
